1.定期觀察細胞培養(yǎng)過程中集落的形成。

2.顯微鏡下計數(shù)大于50個細胞克隆數(shù)，然后按下式計算集落形成率：

人IL-1 alpha/IL1A/IL1F1蛋白注意事項

1.瓊脂對熱和酸不穩(wěn)定，如果反復(fù)加熱，容易降解，產(chǎn)生毒性，同時瓊脂硬度下降。故瓊脂高壓滅菌( 10磅 15分鐘)后按一次用量進行分裝。

2.細胞懸液中，細胞分散度>95%。

3.軟瓊脂培養(yǎng)時，注意瓊脂與細胞混合時溫度不要超過 40℃ ，以免燙傷細胞。

4.接種細胞密度不宜過高。

5.人IL-1 alpha/IL1A/IL1F1蛋白在低密度條件下培養(yǎng)，生存率明顯下降，無限細胞系和腫瘤細胞株克隆形成率一般在10%以上。但初代培養(yǎng)細胞和有限細胞系僅為0.5~5%,甚至為零。為提高集落形成率，必要時在培養(yǎng)基中添加胰島素、*等促細胞克隆形成物質(zhì)。
